Pegula was born to Kim Pegula and Terry Pegula who are the multi-billionaire owners of the Buffalo Bills of the National Football League and the Buffalo Sabres of the National Hockey League.

Pegula and Taylor Townsend competed in the main draw of the US Open doubles competition on August 30, 2011, after receiving a wildcard. Finally, they were defeated by the third-seeded combination of Vania King and Yaroslava Shvedova in the third round.
Pegula received a wildcard to the Indian Wells qualifying round in March 2012. She shocked top-ranked players Bojana Jovanovski and Paula Ormaechea to advance to the main draw, where she was defeated in three sets by Magdaléna Rybáriková.

In Doha, she and Coco Gauff defeated third seeds Elise Mertens and Veronika Kudermetova to win their second doubles championship (and first at the WTA 1000 level). As a consequence, on February 28, 2022, she moved up to No. 29 in the doubles rankings.
Pegula only managed one set in three matches when making her WTA Finals debut, losing all of her matches in the group round. She defeated Yulia Putintseva, Jasmine Paolini, and seventh seed Coco Gauff in Montreal to get to her third consecutive Canadian Open quarterfinal.
Pegula played in the United Cup’s first competition to start her season. Petra Kvitová won her initial match in Group C, but she recovered to overcome Laura Siegmund.
Pegula was one of the favorites to win the Australian Open after her victory over Witek and her consistency during the 2022 season.
Pegula made it to the Qatar Ladies Open finals in both singles and doubles in February. Pegula competed in her first WTA 500 singles final.
