Gay worked in the political science department at Stanford University from 2000 to 2006 as an assistant professor before being promoted to associate professor. Gay worked at the Center for Advanced Study in the Behavioral Sciences as a fellow for the 2003–2004 school year.
In 2007, Gay was named a professor of African-American studies at Harvard, having been recruited in 2006 as a professor of government.

The COVID-19 pandemic of 2020 caused financial and instructional problems for the university. The Faculty of Arts and Sciences recorded losses of $15.8 million for the 2020 fiscal year.
Gay declared in 2021 that the FAS’s primary academic obligations were costing more than it was making. He then started a series of cost-cutting measures, such as freezing salary increases and bonuses for faculty members as well as faculty searches.
Compared to the estimated loss of $112 million, the FAS reported a surplus of $51 million in 2021.
Harvard President Lawrence Bacow stated in June 2022 that he would leave the position in a year. Gay was chosen to succeed Bacow by a search committee headed by Penny Pritzker, Senior Fellow of the Harvard Corporation after 600 nominations were examined.
Harvard University declared on December 15, 2022, that Gay has been chosen to serve as the university’s thirty-first president.
Apart from her education at Stanford and Harvard, Gay was a trustee at Phillips Exeter Academy from 2017 to 2014 and the vice president of the Midwest Political Science Association from 2014 to 2017.
On July 1, 2023, Gay was elected as the first Black president of Harvard University. In addition, she holds the Wilbur A. Cowett Professorship in African and African-American Studies and Government.
